Heat stress and genotype affect the glutenin particles of the glutenin macropolymer-gel fraction

چکیده
Genetic and environmental factors that affect the formation of glutenin macropolymer (GMP) particles were investigated by growing Lance 5C 10, Lance 2C12, Warigal 5C10 and Warigal 2C12 lines under widely differing temperature regimes in greenhouse conditions. The GMP characteristics and mixing properties of flour extracted from mature wheat kernels were systematically studied. The amount of GMP is more sensitive to growing conditions than protein quantity. With the exception of severe heat stress conditions, 5C10 varieties produce larger glutenin particles than 2C12 varieties. The HMW/LMW ratio of GMP is lowered by heat stress, but glutenin particles become larger. In heat stress effects on dough mixing properties, glutenin particle size is more relevant than GMP quantity for dough development time; and GMP quantity is more relevant for band-width at peak resistance. A hyperaggregation model is used to explain how heat stress controls glutenin particle formation. q 2005 Elsevier Ltd. All rights reserved.

The glutenin macropolymer of wheat ̄our doughs: structure±function perspectives
Gluten is the main functional component of wheat, and is the main source of the viscoelastic properties in a dough. Dough properties are primarily governed by the structure of gluten, and interactions within the protein complex. While starch and water are the main components of doughs, the physical properties of a dough arise from interactions between gluten proteins, particularly the disul®de-...

Recently we reported that removal of noncovalently bonded protein from glutenin resulted in decreased viscoelasticity. In that study, two glutenin fractions were isolated by successive gel filtration of glutenin on Sephadex G-200 in the solvents acetic acid-guanidine hydrochloride (GuHCl)-cetyltrimethylammonium bromide (AGC) and sodium dodecyl sulfate (SDS). This note reports the S20,W values o...
متن کاملGluten hydrolysis and depolymerization during sourdough fermentation.
Hydrolysis and depolymerization of gluten proteins during sourdough fermentation were determined. Neutral and acidified doughs in which microbial growth and metabolism were inhibited were used as controls to take into account the proteolytic activity of cereal enzymes. Doughs were characterized with respect to cell counts, pH, and amino nitrogen concentrations as well as the quantity and size d...
متن کاملIsolation and Characterization of Gliadin-Like Subunits from Glutenin
The polypeptide subunits of wheat glutenin obtained after reductive cleavage of disulfide bonds have been separated into two fractions by a modified Osborne technique. Approximately 62% of the subunits by weight are soluble, as is gliadin, in neutral 70% ethanol; the ethanol-soluble subunits are mainly of 44,000 molecular weight (MW). The ethanol-insoluble glutenin subunit fraction is markedly ...
